Nitroso-redox balance in the cardiovascular system.

چکیده

The results with the combination therapy were markedly positive, meeting the composite end point of the trial, which included death from any cause, a first hospitalization for heart failure, and quality-oflife measures. In addition, there was an impressive reduction in the rate of death from any cause of approximately 45 percent, leading to an early termination of the study. This trial raises fascinating issues regarding heart-failure therapeutics and variations in drug susceptibility among populations. Attempting to understand these issues requires insight into the pharmacologic and biologic underpinnings of any drug combination that may enhance nitric oxide availability. This regimen of isosorbide dinitrate and hydralazine (which are available individually as inexpensive, generic formulations), initially selected for its vasodilatory properties in the Veterans Administration Heart Failure Trials (V-Heft) I and II, 2

Role of xanthine oxidoreductase in cardiac nitroso-redox imbalance.

Emerging evidence supports the importance of nitroso-redox balance in the cardiovascular system. Xanthine oxidoreductase (XOR) is a major oxidative enzyme and increased XOR activity, leading to both increased production of reactive oxygen species and uric acid, is implicated in heart failure. Within the heart, XOR activity stimulates cardiomyocyte hypertrophy, apoptosis, and impairs matrix stru...

Nitric oxide synthases, S-nitrosylation and cardiovascular health: From molecular mechanisms to therapeutic opportunities (Review)

The understanding of nitric oxide (NO) signaling has grown substantially since the identification of endothelial derived relaxing factor (EDRF). NO has emerged as a ubiquitous signaling molecule involved in diverse physiological and pathological processes. Perhaps the most significant function, independent of EDRF, is that of NO signaling mediated locally in signaling modules rather than relyin...

A Numerical Simulation of Vanadium Redox Flow Batteries

The recent penetration of renewable sources in the energy system caused a transformation of the needs of the distribution system and amplified the need of energy storage systems to properly balance the electricity grid. Among electrochemical energy storage devices, all vanadium flow batteries are those of the most promising technologies due to their high efficiency, long lifetime, reliability a...

Tetrahydrobiopterin and cardiovascular disease.

Tetrahydrobiopterin (BH4) is an essential cofactor for the aromatic amino acid hydroxylases, which are essential in the formation of neurotransmitters, and for nitric oxide synthase. It is presently used clinically to treat some forms of phenylketonuria (PKU) that can be ameliorated by BH4 supplementation. Recent evidence supports potential cardiovascular benefits from BH4 replacement for the t...

Nitroso - redox balance in control of coronary vasomotor tone 1 2 3

29 Reactive oxygen species (ROS) are essential in vascular homeostasis but may contribute to vascular 30 dysfunction when excessively produced. Superoxide anion (O2) can directly affect vascular tone by 31 reacting with K channels and indirectly by reacting with nitric oxide (NO) thereby scavenging NO and 32 causing nitroso-redox imbalance. After myocardial infarction (MI), oxidative stress inc...
